Polynucleotides represent one of the most significant advances in injectable skin medicine in recent years — a treatment that works at the structural foundation of the skin, rebuilding the extracellular matrix and stimulating collagen synthesis from within.

What Are Polynucleotides?

Polynucleotides (PDRN — Polydeoxyribonucleotide) are highly purified DNA fragments derived from salmon sperm, which shares a remarkable structural similarity with human DNA, minimising any risk of immunological reaction. When injected into the dermis, these DNA chains are recognised by cells as building blocks and signalling molecules, stimulating fibroblast activity, collagen synthesis, and the production of hyaluronic acid within the extracellular matrix.

Unlike hyaluronic acid fillers or skin boosters which introduce volume from outside, polynucleotides instruct the skin to rebuild itself from within. The result is a genuine improvement in the structural quality of the dermis — greater firmness, elasticity, hydration, and resilience — that develops progressively over six to twelve weeks.

“Polynucleotides are one of the treatments we reach for most often for patients concerned about skin quality, laxity, and fine lines — because they address the root cause rather than adding volume on top of depleted tissue. The results are subtle, natural, and lasting.”PHP Aesthetic-Wellness, 22 Harley Street

What Polynucleotides Treat

Skin Laxity & Firmness
By stimulating fibroblasts to produce new collagen and elastin, polynucleotides improve the structural integrity of lax, crepey, or thinning skin — particularly effective on the face, neck, décolleté, and hands.
Fine Lines & Wrinkles
Fine dehydration lines and early wrinkles caused by collagen loss respond well to a course of polynucleotides, which rebuild the dermal scaffolding that supports smooth, plump skin surface.
Under-Eye Hollows
The periorbital area — where skin is thin and tissue depleted — responds particularly well to polynucleotides. Improving hydration and structural quality in this area reduces dark circles and crepey texture without the risk of Tyndall effect associated with hyaluronic acid fillers.
Pigmentation & Skin Quality
By improving barrier integrity and overall dermal health, polynucleotides create the conditions in which existing pigmentation fades more effectively and skin tone becomes more even over time.
Eczema & Barrier Repair
Used during remission periods, polynucleotides strengthen the chronically compromised dermal barrier in eczema-prone skin, reducing flare frequency and improving the skin’s tolerance to environmental triggers.
Hair Loss Support
Injected into the scalp, polynucleotides improve the dermal environment around hair follicles, supporting the results of PRP treatment and improving scalp hydration and tissue quality.

Benefits

Stimulates natural collagen and elastin synthesis — structural improvement, not surface volume
Rebuilds the extracellular matrix for lasting dermal quality improvement
Antioxidant properties protect against free radical damage
Improves skin hydration by stimulating natural hyaluronic acid production
Reduces fine lines and skin laxity progressively over 6–12 weeks
Safe for all skin tones including Fitzpatrick types IV–VI
Minimal downtime — most patients return to normal activities within 24–48 hours
Well tolerated — very low risk of adverse reaction due to structural similarity with human DNA

Treatment Protocol

📋

Initial Course

Three to four sessions spaced two to four weeks apart. The full result of the initial course is assessed at six to eight weeks following the final session, as the biological response to polynucleotide stimulation continues to develop after treatment ends.

Maintenance

A single maintenance session every four to six months sustains the structural improvements achieved and prevents the gradual resumption of collagen loss. Many patients combine maintenance polynucleotide sessions with skin booster or mesotherapy appointments.

Frequently Asked Questions

Are polynucleotides the same as dermal fillers?
No. Dermal fillers add volume using hyaluronic acid gel to restore structure and contour. Polynucleotides stimulate the skin to produce its own collagen and structural proteins — they do not add volume but improve the quality and resilience of the existing tissue. The two treatments address different aspects of ageing and are often used in combination.
How soon will I see results?
Results develop progressively over six to twelve weeks as the stimulated collagen synthesis response matures. Some patients notice improved hydration and skin quality within two to three weeks; the full structural benefit of a course is typically apparent at eight to twelve weeks following the final session.
Is the treatment suitable for sensitive or reactive skin?
Yes. Polynucleotides are very well tolerated across all skin types, including sensitive and reactive skin. They have anti-inflammatory properties and a very low risk of adverse reaction. They are one of the treatments we use most frequently for eczema-prone and rosacea-affected skin during stable periods.
Are polynucleotides suitable for darker skin tones?
Yes — polynucleotides carry no pigmentation risk and are appropriate for all Fitzpatrick skin types. By improving barrier integrity and overall dermal health, they can be used as part of a pigmentation management protocol in medium-to-deep skin tones.
What is the difference between polynucleotides and skin boosters?
Skin boosters deliver hyaluronic acid directly into the dermis for immediate deep hydration and radiance. Polynucleotides stimulate the skin to produce its own structural proteins and hyaluronic acid, improving quality from within. Both are valuable — skin boosters work faster; polynucleotides produce more lasting structural change. Most patients benefit from both.
